run Chkdsk diagnostic.
Start->Accessories->Command Prompt
At the command prompt, type the following command:
chkdsk c: /F/R
Hit return
The operating system will tell you it cannot do this now and must reboot to perform the chkdsk. Say "y" for yes and then reboot the computer.
The computer will launch into chkdsk and go through 5 LONG and TEDIUOUS checks. The larger the hard disk, the longer it will take. It may take 2 - 3 hours, but keep your hands OFF the computer and let it complete the entire diagnostic.
It will find damaged clusters and sectors and will mark the damages as unusable and that should fix the problem so defrag can complete. I am guessing you just have some ware and tear on that hard disk and now there are problems holding it up.
